My sister and her husband are getting a divorce. They are currently separated and there son lives wiht her 6 days a week and him one. She works massive weeks and puts her son into day care coz the husband has no interest in taking him. He works 3 nights a week for 4 hours a shift. There house is on the market, which is in my sisters name, she is paying off the mortgage, while he lives there rent free. She also paid for his car. We saw an email the other day that accidentally got sent to her from his mother saying that as its the family home and they have joint custody he is entitled to half of everything no matter who paid for it all. Is this true?

I believe 50/50 is only in 10 years of marrage.
Why wouldn’t he be entitled to a 50-50 split? If the house belonged to your sister before she got married, then she will probably get it, but generally assets obtained during the course of the marriage will be split 50-50.
If they genuinely share caring for the son, or if custody is awarded to the husband, then your sister may also be required to pay child support.
